Principle of test
Chyle tests are often used to diagnose chyluria, etc. Chyluria is a chylous turbid urine composed of fat particles and proteins. Chyluria becomes clear after extraction of chyluria particles or fat droplets in urine with fat-soluble organic solvent (ethyl ether) is a positive chyluria test. Fat droplets in chyluria can be colored with a fat-soluble dye (Sudan III) and identified under a microscope.

Instruments and reagents
Microscope, Centrifuge, sample, ethyl ether, Sudan III acetic acid ethanol stain.
Test steps
- Add 5ml sample in tub
- Add in 2ml of Ethyl ether
- Mix and shake
- Leave for a few minutes
- Centrifuge
- Take the interface layer of inhaled ether and urine Smear, add 1 drop of Sudan III stain solution
- Examine under microscope
Determination of results
Check if there are red fat droplets.
Safety risk of ethyl ether :
- Extremely flammable liquid and vapor.
- Harmful if swallowed. May cause drowsiness or dizziness. Long-term low concentration inhalation may cause headache, dizziness, fatigue, lethargy, urinary protein and other symptoms. Repeated exposure may cause skin dryness or cracking.
- The after-effects of acute exposure include headache, irritability or depression, salivation, vomiting, decreased appetite and hyperhidrosis. Liquids or high concentrations of vapor are irritating to the eyes.
